目的建立血小板捐献者HLA基因分型资料库,探讨HLA-A、B基因频率及单体型频率,为临床患者提供HLA匹配的血小板。方法应用聚合酶链反应-序列特异性引物方法和Luminex-SSO方法对730名沈阳地区血小板捐献者进行中低分辨率HLA-A、B基因分型,根据HLA表型群体资料,使用最大数学预期值算法(Expectation-Maximization,EM)计算HLA-A、B座位单体型频率。结果 HLA-A位点共检出15种等位基因,HLA-B位点共检出36种等位基因,单体型数222种。结论为向临床提供HLA匹配的血小板,各地区有必要建立血小板捐献者HLA基因分型资料库。
Objective To establish a HLA genotyping database of platelet donors, to explore HLA-A and B gene frequencies and haplotype frequencies, and to provide HLA-matched platelets to clinical patients. Methods The low-resolution HLA-A and B genotypes of 730 platelet donors in Shenyang were genotyped by polymerase chain reaction-sequence-specific primers and Luminex-SSO method. According to HLA phenotypic population data, the maximal mathematical expectation The value algorithm (Expectation-Maximization, EM) HLA-A, B seat haplotype frequency. Results Fifteen alleles were detected at HLA-A locus, 36 alleles at HLA-B locus and 222 haplotypes. Conclusion In order to provide HLA-matched platelets clinically, it is necessary to establish HLA genotyping database for platelet donors in all regions.
