目的探讨灯盏花素对家兔心肌缺血再灌注损伤(MIRI)的保护作用。方法将19只家兔随机分为假手术组(6只)、模型组(7只)、治疗组(6只)。以穿线结扎左冠状动脉前降支制备大鼠心肌缺血/灌注模型,治疗组于结扎前5min于耳缘静脉注射灯盏花素,观察各组心肌超微结构,右室血清乳酸(LA)、游离脂肪酸(NEFA)和心肌组织中LA、NEFA、三磷酸腺苷(ATP)水平的变化。结果与假手术组比较,模型组血清及心肌组织中LA、NEFA水平明显升高(P<0.05或P<0.01),心肌组织中ATP含量显著降低(P<0.01),心肌超微结构损害明显;与模型组比较,治疗组血清及心肌组织中LA、NEFA水平明显降低(P<0.05或P<0.01),心肌组织中ATP含量显著增高(P<0.01),心肌超微结构损害减轻。结论灯盏花素可通过改善心肌能量代谢障碍和保护线粒体结构对MIRI起到保护作用。
Objective To investigate the protective effect of breviscapine on myocardial ischemia-reperfusion injury (MIRI) in rabbits. Methods Nineteen rabbits were randomly divided into sham operation group (n = 6), model group (n = 7) and treatment group (n = 6). The model of myocardial ischemia / perfusion was established by ligating the left anterior descending artery of the left anterior descending coronary artery. The treatment group was given breviscapine 5 minutes before ligation to observe the ultrastructure of the myocardium, the content of lactic acid (LA) Free fatty acid (NEFA) and myocardial tissue LA, NEFA, ATP levels. Results Compared with sham operation group, LA and NEFA levels in serum and myocardium of model group were significantly increased (P <0.05 or P <0.01), ATP content in myocardial tissue was significantly decreased (P <0.01), myocardial ultrastructure damage was obvious Compared with the model group, the levels of LA and NEFA in the serum and myocardium of the treated group were significantly decreased (P <0.05 or P <0.01), the content of ATP in the myocardium was significantly increased (P <0.01), and the myocardial ultrastructure damage was alleviated. Conclusion Breviscapine can protect MIRI by improving myocardial energy metabolism and protecting mitochondrial structure.
