目的 :探讨细胞间粘附分子 1(ICAM 1)在大鼠视网膜缺血再灌注损伤中表达的动态变化及N 乙酰半胱氨酸(NAC)的保护作用。方法 :建立大鼠视网膜缺血再灌注模型 ,6 0只SD大鼠随机分为缺血再灌注组和缺血再灌注 +NAC治疗组。每组按再灌注后不同时间段分为 1、6、12、2 4、4 8、72小时组 ,每组 5只 ,以原位杂交法及免疫组织化学法检测不同时期视网膜中ICAM 1的表达 ,每只大鼠处死前行视网膜电图 (ERG)检测。结果 :缺血再灌注组在 6小时开始检测到ICAM 1mRNA的表达 ,在 2 4小时表达最强 ,以后逐渐减弱。在 12小时才能检测到ICAM 1蛋白的表达 ,2 4小时表达最强 ,缺血再灌注 +NAC治疗组在再灌注 6小时亦能检测到ICAM 1mRNA的表达 ,在 12小时能检测到ICAM 1蛋白的表达 ,但低于同期缺血再灌注组的表达水平 (P <0 0 5 )。缺血再灌注组在再灌注 6小时后各期ERGb波振幅明显低于同期缺血再灌注 +NAC治疗组(P <0 0 5 )。结论 :视网膜缺血再灌注损伤时ICAM 1参与其病理损伤过程 ,NAC可通过抑制ICAM 1的表达而减轻视网膜缺血再灌注损伤。
Objective: To investigate the dynamic changes of intracellular adhesion molecule 1 (ICAM 1) expression in retinal ischemia-reperfusion injury and the protective effect of N-acetylcysteine (NAC). Methods: The rat model of retinal ischemia-reperfusion was established. Sixty SD rats were randomly divided into ischemia-reperfusion group and ischemia-reperfusion + NAC group. The rats in each group were divided into 1, 6, 12, 2, 4, 8, 8 hours and 2 hours after reperfusion. Five rabbits in each group were tested for in situ hybridization and immunohistochemistry Expression, each rat was sacrificed before the electroporation (ERG) test. Results: ICAM 1 mRNA expression was detected at 6 hours after ischemia-reperfusion in rats, which was the strongest expression at 24 hours and then decreased gradually. The expression of ICAM 1 protein was detected at 12 hours and the expression was highest at 24 hours. ICAM 1 mRNA was also detected at 6 hours after reperfusion in ischemia-reperfusion + NAC group, and ICAM 1 protein was detected at 12 hours But lower than that of ischemia-reperfusion group (P <0.05). The amplitude of ERGb wave in each group was significantly lower than that in ischemia / reperfusion + NAC group (P <0.05) at 6 h after reperfusion. CONCLUSION: ICAM-1 is involved in the process of retinal ischemia-reperfusion injury. NAC can reduce the damage of retinal ischemia-reperfusion injury by inhibiting the expression of ICAM-1.
