大鼠急性CCl_4损伤肝库普弗细胞对肝星状细胞增殖和Ⅰ型胶原分泌的影响

通过细胞体外培养观察急性CCl4 肝损伤的大鼠肝库普弗细胞对正常肝星状细胞的激活作用。方法 以 5 0 %CCl4 橄榄油一次灌胃复制大鼠急性肝损伤模型 ,分离模型大鼠肝库普弗细胞并原代培养 ,收集条件培养液 ,以其温育正常大鼠分离的原代培养肝星状细胞 ,二苯基四氮唑溴盐 (MTT)法测定细胞增殖 ,ELISA法测定Ⅰ型胶原分泌。结果 损伤肝库普弗细胞条件培养液可显著促进肝星状细胞的增殖及Ⅰ型胶原分泌 ,并具一定量效、时效关系 ;还可促进星状细胞表型向肌成纤维样细胞转化 ,而正常大鼠分离的肝库普弗细胞则无明显作用。结论 肝损伤时库普弗细胞对肝星状细胞的活化有多方面旁分泌激活作用 The activation of normal hepatic stellate cells was observed in vitro by CCl4-induced hepatotoxicity in vitro. Methods Acute liver injury model was induced by orally gavage with 50% CCl4 olive oil. The rat liver Pupu cells were isolated and cultured in primary culture. The conditioned medium was collected and incubated in primary culture of normal rats Hepatic stellate cells and diphenyltetrazolium bromide (MTT) method were used to measure cell proliferation. ELISA was used to determine the type I collagen secretion. Results Injury of liver Kupffer cells conditioned medium could significantly promote the proliferation of hepatic stellate cells and the secretion of type I collagen with a certain dose-effect and time-dependent relationship. It also promoted the transformation of stellate cell phenotype into myofibroblast- The normal rat liver Pupu cells isolated from no significant effect. Conclusion Kupffer cells have many roles in paracrine activation of hepatic stellate cells in liver injury
