目的:观察丁酸钠对结肠癌细胞株SW480的生长抑制、凋亡情况以及对生成素(Survivin)表达水平的影响,探讨其预防结肠癌的作用机制。方法:人传代结肠癌细胞株SW480经不同浓度丁酸钠处理后,在不同时段收集细胞,分别用四唑蓝法检测肿瘤细胞生长增殖率,流式细胞仪检测细胞周期和凋亡变化,免疫细胞化学技术观察对Survivin基因和血管内皮生长因子(VEGF)表达水平。结果:随着培养液中丁酸浓度的不断升高和培养时间的延长,SW480细胞的生长逐渐受到抑制。G1期细胞明显增多,S期细胞明显减少,细胞凋亡率逐渐增高。Sur-vivin基因和VEGF随着丁酸浓度的不断升高而表达下降。结论:丁酸钠能抑制SW480细胞株增生,诱导凋亡,并抑制Survivin基因和VEGF表达。
OBJECTIVE: To observe the effect of sodium butyrate on the growth inhibition, apoptosis and the expression of Survivin in colon cancer cell line SW480 and to explore its mechanism of action in preventing colon cancer. Methods: Human colorectal cancer cell line SW480 was treated with different concentrations of sodium butyrate. The cells were harvested at different time points. The growth and proliferation of tumor cells were detected by tetrazolium method. The cell cycle and apoptosis were detected by flow cytometry. Cytochemical techniques were used to observe the expression of survivin gene and vascular endothelial growth factor (VEGF). Results: The growth of SW480 cells was inhibited gradually with the increase of the concentration of butyric acid in culture medium and the prolongation of culture time. G1 phase cells increased significantly, S phase cells decreased significantly, the rate of apoptosis increased gradually. Survivin gene and VEGF decreased with the increasing concentration of butyrate. CONCLUSION: Sodium butyrate can inhibit the proliferation of SW480 cell line, induce apoptosis and inhibit the expression of Survivin gene and VEGF.
