本文报道用~3H-哌唑嗪、~3H-育亨宾为放射配体,建立测定大鼠肝脏的α_1、α_2受体的方法。实验结果得到良好的饱和曲线,重复性良好,用Scatchard作图法计算出受体数(R_T)及亲和力(以解离常数K_d表示)。大鼠肝α_1受体R_T值为74.8±4.5fmol/mg蛋白((?)±s),K_d值为0.37±0.07nmol/L;α_2受体R_T值为21.4±3.2fmol/mg蛋白,K_d值为0.37±0.07nmol/L。本实验数据稳定,重复性好,是测定肝脏α-肾上腺素受体一种较好的方法。
This paper reports the use of ~ 3H-prazosin, ~ 3H-yohimbine as a radioligand, the establishment of the determination of rat liver α_1, α_2 receptor method. The experimental results obtained a good saturation curve, good repeatability, using Scatchard plot method to calculate the number of receptors (R_T) and affinity (dissociation constant K_d said). The R_T value of rat liver α_1 receptor was 74.8 ± 4.5 fmol / mg protein (± s), the K_d value was 0.37 ± 0.07 nmol / L, the R_T value of α_2 receptor was 21.4 ± 3.2 fmol / mg protein, K_d value 0.37 ± 0.07 nmol / L. The experimental data is stable and reproducible, is a good method for the determination of hepatic α-adrenoceptor.
