本研究以新生大鼠心肌组织为研究对象,分别采用组织学、免疫组织化学及甲苯胺蓝染色方法,对新生大鼠心肌组织中telocytes形态特征和免疫标志进行观察与分析。其中,组织学染色与甲苯胺蓝染色结果显示,新生大鼠心肌组织中存在大量间质细胞,其中telocytes的分布多紧贴于心肌纤维束周围或纤维之间,细胞呈现长梭形、纺锤形及三角形等多种形态,胞质少、有一至多条长而细的细胞突起,胞核结构较致密、着色较深。免疫组化染色结果进一步证实新生大鼠心肌组织中其不但表达间质细胞的免疫标志vimentin,有些还表达c-kit/CD117和CD34两种与干细胞相关的表面标志物。本研究从组织形态学的角度阐明了telocytes的基本特征和分布,为进一步揭示其功能奠定了在体形态结构上的基础。
In this study, the myocardial tissue of newborn rats was taken as the research object. The morphology and immune sign of telocytes in neonatal rat myocardium were observed and analyzed by histology, immunohistochemistry and toluidine blue staining respectively. Among them, histological staining and toluidine blue staining showed that a large number of interstitial cells existed in the neonatal rat myocardium. Among them, the distribution of telocytes was close to or around the myocardial fiber bundles, And triangles and other forms, less cytoplasm, there are more than one long and thin cell protrusions, the nucleus structure is more dense, darker. Immunohistochemical staining further confirmed vimentin, an immunological marker of interstitial cells in neonatal rat myocardium, and some also expressed stem cell-associated surface markers of c-kit / CD117 and CD34. This study elucidates the basic characteristics and distribution of telocytes from the perspective of histomorphology, which lays the foundation for the morphological structure of the telocytes in order to further reveal its function.
