空中子囊孢子捕捉是目前小麦赤霉病测报中菌量动态监测的有效方法之一,常用方法有电动孢子捕捉法和水平琼脂培养法2种。前法捕捉效率高,反应灵敏,但镜检费时费力,漏查率高,特别是无法辨清孢子的死活;琼脂培养法,虽能反映空中有效菌量,但缺点是捕捉效率太低,灵敏度差,且易受气候影响。为了发挥电动捕捉法的长处,克服其缺点,我们进行了赤霉菌手囊孢子死活的染色鉴定,取得预期效果。现简报如下: 方法 1.染色液的配制:选用中性红和次甲基蓝,分别配成0.2%等容积的水溶液,为了加速次甲基蓝的溶解,将溶液加温至45℃左右,并搅拌。待两者充分溶解后,将中性红倒入次甲基蓝溶液中。并搅拌后装入试剂瓶中。
