目的研究肝刺激物(hepatic stimulator substance,HSS)对人肝癌细胞凋亡的影响。方法 用无血清培养人肝癌细胞诱导典型的细胞凋亡,以此为模型应用流式细胞术及流式细胞免疫荧光技术,检测HSS作用不同时间后,对无血清培养人肝癌细胞诱导凋亡比率的影响,并观察HSS对常规培养人肝癌细胞的P53蛋白表达变化的影响。结果HSS作用于无血清培养的人肝癌细胞,细胞凋亡比率随时间延长而增加;HSS作用于常规培养的人肝癌细胞,细胞 P53蛋白的表达随时间延长而减弱。结论 HSS对无血清培养人肝癌细胞凋亡有促进作用。
Objective To study the effect of hepatic stimulator substance (HSS) on the apoptosis of human hepatoma cells. Methods Human hepatocellular carcinoma cells were cultured in serum-free medium to induce typical apoptosis. Flow cytometry and flow cytometry were used as the model to detect the effect of HSS on the induction rate of apoptosis in serum-free human hepatoma cells , And observe the effect of HSS on the expression of P53 protein in routinely cultured human hepatoma cells. Results HSS was applied to human hepatocarcinoma cells in serum-free medium. The apoptosis rate increased with time. The effect of HSS on routine cultured human hepatocellular carcinoma cells showed that the expression of P53 protein decreased with time. Conclusion HSS can promote the apoptosis of human hepatoma cells in serum-free medium.
