用灵敏的过氧化物酶免疫“夹心”技术,发现健康人AB型血清中含有一种抗乳腺癌细胞的肿瘤相关抗体(TAA)。用这种血清处理福尔马林固定石蜡包埋的组织切片,检查了16例良性和13例恶性的乳腺病,只有恶性上皮细胞显示与这种TAA有特异的结合,而良性或正常组织则不与TAA结合。经显色后可在同一张切片上明显的区分出乳腺的良恶性细胞。对此种TAA的本质进行了比较研究,发现主要是在AB型血清的免疫球蛋白Igm成分中。经IgM处理的13例恶性乳腺病的组织切片中有12例呈中到高强度染色,只有1例染色较弱,但也可肯定是阳性染色。而经AB型血清中的IgA和IgG处理后,染色强度比IgM处理的同样的切片要弱得多,用IgD处理的切片染色则只呈痕迹量甚至测不出来。另外,其中8
Using a sensitive peroxidase-immunized “sandwich” technique, healthy human AB serum was found to contain a tumor-associated antibody (TAA) against breast cancer cells. Formalin-fixed paraffin-embedded tissue sections were treated with this serum and 16 benign and 13 malignant breast diseases were examined. Only malignant epithelial cells showed specific binding to this TAA, whereas benign or normal tissues were Not combined with TAA.
