以明胶为载体,液体石蜡为油相,Span80为乳化剂,戊二醛为交联剂,采用乳化交联法制备芦丁明胶微球,通过单因素实验和正交设计实验筛选出最佳工艺条件,并对工艺重现性进行了考察。利用扫描电镜、红外光谱、X衍射图谱等手段对产物进行了表征,计算出明胶微球的载药量和包封率。结果表明:芦丁用量及交联剂用量是影响微球性能的主要因素;微球形态圆整、表面光滑、大小均匀,其粒径为5~20μm,载药量为3.55%,包封率为94.5%。
Using gelatin as carrier, liquid paraffin as oil phase, Span80 as emulsifier and glutaraldehyde as cross-linking agent, rutin gelatin microspheres were prepared by emulsion cross-linking method. The best technology was selected by single factor experiment and orthogonal design experiment Conditions, and process reproducibility were investigated. The products were characterized by scanning electron microscopy, infrared spectroscopy and X-ray diffraction. The drug loading and entrapment efficiency of gelatin microspheres were calculated. The results showed that the dosage of rutin and the amount of crosslinking agent were the main factors affecting the microsphere properties. The morphology of the microspheres was round, smooth and uniform in size. The diameter of the microspheres was 5 ~ 20μm and the drug loading was 3.55% 94.5%.
