目的 :探讨补阳还五汤一次和连续灌服给药对大鼠实验性动脉血栓形成前后动脉血中血小板活化因子 (PAF)含量的影响。方法 :将雄性Wistar大鼠分别随机分为 3组。用药组一次或连续 (1 4天 ,每天 1次 )灌服补阳还五汤水煎液 (3 2 g/kg、1 6g/kg) ;对照组一次或连续 (1 4天 ,每日 1次 )灌服地塞米松片溶液 (1mg/kg、0 5mg/kg) ;空白组均灌服等体积蒸馏水(1 0ml/kg)。末次给药 2h后 ,复制实验性动脉血栓模型 ,用PAF含量的生物测定法 ,测定血栓形成前后动脉血中PAF含量和血栓OT值、血栓干重及血栓 -体重指数。结果 :补阳还五汤一次给药对血栓形成前后动脉血中PAF含量和血栓OT值无明显影响 ;而连续给药能明显降低血栓形成后动脉血中PAF含量 (P <0 0 5 ) ,并明显提高动脉血栓OT值 (P <0 0 5 ) ,降低血栓干重和血栓 -体重指数 (P <0 0 5 )。结论 :补阳还五汤对病理状态下PAF代谢有一定的拮抗效应 ,并对动脉血栓的发生、发展有一定的抑制作用。
Objective: To investigate the effects of once-and-continuous administration of Buyang Huanwu Decoction on arterial blood platelet activating factor (PAF) content before and after experimental arterial thrombosis in rats. Methods: Male Wistar rats were randomly divided into 3 groups. The treatment group was fed once or continuously (1 day, 1 time per day) with decoction of Buyang Huanwu Decoction (3 2 g/kg, 1 6g/kg); the control group was once or continuously (1 4 days, 1 time per day). ) The dexamethasone tablet solution (1 mg/kg, 0 5 mg/kg) was given to the rats; the blank group was given an equal volume of distilled water (10 ml/kg). Two hours after the last administration, the experimental arterial thrombosis model was duplicated, and the PAF content and thrombus OT value, thrombus dry weight, and thrombus-body mass index of arterial blood before and after thrombus formation were determined by bioassay of PAF content. Results: One dose of Buyang Huanwu Decoction had no significant effect on the content of PAF and thrombus thrombus in arterial blood before and after thrombosis, while continuous administration could significantly reduce the content of PAF in arterial blood after thrombosis (P < 0.05). And significantly increase the arterial thrombus OT value (P <0 05), reduce the thrombus dry weight and thrombus - body mass index (P <0 05). Conclusion: Buyang Huanwu decoction has a certain antagonistic effect on PAF metabolism under pathological conditions, and has a certain inhibitory effect on the occurrence and development of arterial thrombosis.
