目的:研究载脂蛋白A-I(ApoA-I)对内毒素血症小鼠肝组织的保护作用。方法:建立内毒素血症小鼠急性肝损伤模型,观察人血浆ApoA-I处理后各组小鼠肝脏的组织学改变,RT-PCR检测小鼠肝脏CD14的表达。结果:内毒素血症肝损伤小鼠肝脏发生了明显坏死,而ApoA-I治疗组小鼠肝脏仅发生轻微坏死;内毒素血症小鼠肝脏组织CD14mRNA表达水平明显增高,ApoA-I治疗后,CD14mRNA表达水平下降(P<0.05)。结论:ApoA-I对内毒素血症小鼠肝损伤具有保护作用,其作用机制可能与ApoA-I降低肝组织LPS受体CD14的过度表达有关。
Objective: To study the protective effect of apolipoprotein A-I (ApoA-I) on liver tissue of endotoxemia mice. Methods: The model of acute liver injury in mice with endotoxemia was established. The histological changes of liver in each group were observed after ApoA-I treatment. The expression of CD14 in liver of mice was detected by RT-PCR. Results: The liver of mice with endotoxemia had obvious necrosis in the liver, but mild necrosis in the liver of mice treated with ApoA-I. The expression of CD14mRNA in the liver of endotoxemia mice was significantly increased. After ApoA-I treatment, CD14 mRNA expression decreased (P <0.05). CONCLUSION: ApoA-I has a protective effect on liver injury induced by endotoxemia in mice, and its mechanism may be related to the decrease of hepatic LPS receptor CD14 overexpression by ApoA-I.
