目的通过检测感染旋毛虫的小鼠膈肌中IL-8受体CXCR1/2的表达以及肌肉组织纤维化程度,探讨G31P对旋毛虫囊包形成的影响以及防治效果。方法 30只雌性BALB/c小鼠随机分成三组:阳性模型对照组(n=12,皮下注射生理盐水),G31P治疗组(n=12,皮下注射G31P)和阴性正常对照组(n=6,皮下注射生理盐水)。阳性模型对照组与G31P治疗组小鼠应用口饲法感染旋毛虫囊包,(100±5)个/只,于感染后第10天皮下注射G31P(500μg/kg),1次/2d,连续14d,阳性对照组和阴性对照组分别皮下注射等量生理盐水。感染24d后无菌解剖小鼠,收取膈肌组织,用于HE染色和免疫组化分析。结果与阳性模型对照组相比,G31P治疗组旋毛虫囊包数量明显减少(P<0.01),而且囊包大多形成不完整、纤维化程度轻。炎症因子受体CXCR1和CXCR2的表达均显著低于模型组(P<0.01)。结论 G31P具有下调膈肌组织中炎症因子IL-8受体CXCR1和CXCR2表达的作用,从而拮抗旋毛虫囊包纤维化的形成,并对旋毛虫幼虫虫体结构的完整性产生一定的影响。
Objective To investigate the effect of G31P on the growth of Trichinella spiralis and its prevention and control by detecting the expression of IL-8 receptor CXCR1 / 2 and the degree of muscle fibrosis in mouse diaphragm infected with Trichinella spiralis. Methods Thirty female BALB / c mice were randomly divided into three groups: positive model control group (n = 12, subcutaneous injection of normal saline), G31P treatment group (n = 12, subcutaneous injection of G31P) and negative control group , Subcutaneous injection of saline). Positive control mice and G31P-treated mice were infected with Trichinella spiralis (100 ± 5) / g only by oral gavage, and injected with G31P (500μg / kg) subcutaneously once daily for 2 days 14d, positive control group and negative control group were injected subcutaneously with normal saline. After 24 days of infection, the mice were aseptically dissected and the diaphragm tissue was collected for HE staining and immunohistochemical analysis. Results Compared with the positive model control group, the number of Trichinella kits in G31P treatment group was significantly decreased (P <0.01), and the majority of capsular formation was incomplete and the degree of fibrosis was light. The expressions of CXCR1 and CXCR2 in inflammatory cytokines were significantly lower than those in model group (P <0.01). Conclusion G31P can down-regulate the expression of IL-8, CXCR1 and CXCR2 in diaphragmatic muscle tissue and thus antagonize the formation of Trichinella spiralis fibrosis and have an impact on the structural integrity of Trichinella spiralis larvae.
