目的:建立茯苓与茯苓皮中茯苓酸含量的测定方法,比较不同栽培条件的茯苓与茯苓皮中茯苓酸的含量。方法:采用反相高效液相色谱法,色谱柱为Diamonsil C18,4.6×250nm,5um);乙腈-0.4%乙酸(85:15)为流动相;流速为1m L/min;检测波长为206nm,柱温25℃。结果 :茯苓酸在0.49ug~5.88ug范围内与其色谱峰面积呈良好线性关系,r=0.9997,平均回收率96.94,RSD=1.97%。结论:该方法简便、专属、稳定,可用于茯苓及茯苓皮中茯苓酸的含量测定。
Objective: To establish a method for the determination of Poria cocos acid in Poria cocos and Poria cocos and compare the content of Poria cocos acid in Poria cocos and Poria cocos under different cultivation conditions. Methods: The mobile phase was Diamonsil C18 (4.6 × 250nm, 5um) with acetonitrile-0.4% acetic acid (85:15) at a flow rate of 1m L / min. The detection wavelength was 206nm, Column temperature 25 ℃. Results: There was a good linear relationship between the content of pachymalin and its peak area in the range of 0.49ug ~ 5.88ug, r = 0.9997, the average recovery 96.94, RSD = 1.97%. Conclusion: The method is simple, specific, stable and can be used for the determination of Poria cocos in Poria and Poria cocos.
