目的 研究健康志愿者单次 po不同剂量国产甲磺酸左氧氟沙星的药动学。 方法 30名健康志愿者分 3组 po 10 0 ,2 0 0 ,30 0mg甲磺酸左氧氟沙星 ,血及尿药浓度均采用HPLC测定。数据用 3P87药动学程序进行模型拟合并计算药动学参数。结果 甲磺酸左氧氟沙星药 时曲线符合二室模型 ,po 10 0 ,2 0 0 ,30 0mg后其主要药动学参数 :tpeak分别为 1.2 87,1.12 1,1.386h ,t1/2 β分别为 7.6 6 9,6 .5 6 9,6 .187h ,AUC分别为 11.418,2 1.5 0 2 ,32 .746mg·h·L-1,0～ 2 4h尿药回收率分别为72 .2 4% ,6 6 .31% ,6 8.6 2 %。主要药动学参数在 3个剂量组之间无显著性差异。结论 国产甲磺酸左氧氟沙星在健康人体内的处置无剂量依赖性。本研究可为该药的临床应用提供一定科学依据
Objective To study the pharmacokinetics of levofloxacin mesylate in healthy volunteers with single dose of po. Methods Thirty healthy volunteers were divided into 3 groups: po 10 0, 2 0 0, 300 mg levofloxacin mesylate, blood and urine concentration were determined by HPLC. Data were modeled using the 3P87 pharmacokinetic program and pharmacokinetic parameters were calculated. Results The pharmacokinetics parameters of levofloxacin mesylate were in accordance with the two-compartment model. The main pharmacokinetic parameters of tricaprylin were 1.2 87,1.12 1,1.386 h after po 10 0, 2 0 0 and 30 0 mg respectively, and t1 / 2 β were 7.6 6 9,6 .5 6 9,6. 187h, the AUC were 11.418, 2 1.5 0 2, 32 .746mg · h · L-1, 0 ~ 2 4h urinary drug recovery rates were 72.24%, 6 6.31%, 6 8.6 2%. The main pharmacokinetic parameters in the three dose groups no significant difference. Conclusion Domestic levofloxacin mesylate treatment in healthy people without dose-dependent manner. This study may provide a scientific basis for the clinical application of this medicine
