目的观察在常压7%氧浓度环境下于不同持续时间大鼠小脑浦肯野细胞组织学及其超微结构的急性损伤病理改变。方法 Wistar大鼠120只,随机分为缺氧组和对照组,雌雄各半,每组60只,其中缺氧组大鼠置入常压低氧舱内,氧浓度为7%,于持续缺氧1、3、5、7、10、14 d与相应对照组分别处死,小脑取材、固定、制片,观察其光镜和电镜下的结构变化,并从每只大鼠小脑组织所制的切片中随机选片,于光镜下自桥脑侧计数500个浦肯野细胞,计算变性细胞的发生率。结果从持续缺氧1~7 d过程中,浦肯野细胞的变性程度呈进行性加重,缺氧性改变细胞的数量呈进行性增加;于持续缺氧10~14 d时,缺氧性改变的程度呈进行性减轻趋势,变性细胞的数量亦呈减少趋势。光镜观察、电镜观察及变性细胞发生率三者结果一致。结论在不同持续常压缺氧过程中大鼠小脑浦肯野细胞的变性程度及其数量呈早期进行性加重和增多—后期渐适应恢复的规律性变化。
Objective To observe the pathological changes of Purkinje cell histology and its ultrastructure in different durations under the condition of 7% oxygen and atmospheric pressure. Methods 120 Wistar rats were randomly divided into hypoxia group and control group, with 60 male and 60 female. The rats in hypoxia group were placed in normoxia chamber at an oxygen concentration of 7% Oxygen was sacrificed on the 1st, 3rd, 5th, 7th, 10th, 14th day respectively in the control group. The cerebellum was taken out, fixed and made into pieces. The structure changes of the cerebellum under light and electron microscope were observed. Results During the first 7 days of hypoxia, the degree of degeneration of Purkinje cells increased progressively and the number of hypoxic cells increased progressively. Conclusions The degree of degeneration of Purkinje cells in rat cerebellum during different durations of hypobaric hypobaric hypoxia and the number of degenerative changes in the cerebellum range from early to late aggravating and regressing.
