目的:探讨茶多酚能否诱导鼻咽癌细胞系 CNE1-LMP1凋亡及凋亡发生的分子机制。方法:用不同浓度的茶多酚处理 CNE1-LMP1细胞,分别用荧光显微镜、 DNA琼脂糖凝胶电泳、 MTT和流式细胞仪等方法,观察细胞生化和形态学改变;同时,确定茶多酚诱导细胞凋亡过程中 caspase-3活性的动力学变化。结果:茶多酚处理后,荧光显微镜下可见 CNE1-LMP1细胞在形态学上出现细胞核固缩、核碎裂、凋亡小体等凋亡细胞的形态学改变;琼脂糖凝胶电泳 DNA呈典型的“梯状”条带; MTT测定发现茶多酚处理后细胞生长受到不同程度的抑制。同时, CNE1-LMP1细胞在凋亡过程中 caspase-3活性的升高,呈明显的时效关系。结论:茶多酚能诱导 CNE1-LMP1细胞凋亡,可能通过 caspase-3的活化诱导细胞凋亡。
Objective: To investigate the molecular mechanism of tea polyphenols inducing apoptosis and apoptosis of nasopharyngeal carcinoma cell line CNE1-LMP1. Methods: CNE1-LMP1 cells were treated with different concentrations of tea polyphenols. The biochemical and morphological changes of CNE1-LMP1 cells were observed by fluorescence microscope, DNA agarose gel electrophoresis, MTT and flow cytometry, respectively. At the same time, Kinetic Changes of Caspase-3 Activity Induced by Apoptosis. Results: After treated with tea polyphenols, morphological changes of CNE1-LMP1 cells were found on the morphology of apoptotic cells such as nuclear condensation, nuclear fragmentation and apoptotic bodies. The DNA of agarose gel electrophoresis showed typical The “ladder-like” bands were detected by MTT assay. Cell growth was inhibited to some extent by the treatment of tea polyphenols. At the same time, the caspase-3 activity in CNE1-LMP1 cells increased during apoptosis, showing a time-dependent relationship. CONCLUSION: Tea polyphenols can induce apoptosis in CNE1-LMP1 cells and induce apoptosis possibly through the activation of caspase-3.
