目的 :探讨白细胞介素 - 6 (IL- 6 )、白细胞介素 - 1(IL- 1)和血小板源生长因子 (PDGF)对大鼠肾小球系膜细胞 (Ms C)增殖的影响。方法 :采用改良的 MTT法和 Brd U掺入法检测 SD大鼠体外 Ms C悬液中加入 IL- 6、IL- 1及 PDGF后 12 h及 2 4h的增殖情况 ,分 7组进行观察。结果 :IL- 6组、IL- 1组和 PDGF组 12 h及 2 4h的光密度 (OD值 )与标记指数 (L I)均比 2 % FCS组高 ;IL - 6加 IL - 1组或 IL - 6加 PDGF组 12 h及 2 4h的 OD值与 L I也均比 2 % FCS组高 ,但所有实验组均不能达到 2 0 % FCS组的细胞增殖水平。联合应用双因子刺激 12 h及 2 4h均未见明显的协同促增殖作用。结论 :IL - 6、IL - 1及 PDGF均能在一定程度上刺激大鼠肾小球 Ms C增生 ,联合应用双因子刺激无明显的协同效应。
AIM: To investigate the effects of interleukin 6 (IL-6), interleukin-1 (IL-1) and platelet-derived growth factor (PDGF) on the proliferation of rat mesangial cells (MsC). Methods: Proliferation of 12 h and 24 h after the addition of IL-6, IL-1 and PDGF in Ms C suspension of SD rats was detected by modified MTT assay and BrdU incorporation assay. Results: The optical density (OD) and labeling index (LI) of IL - 6, IL - 1 and PDGF groups were higher than those of 2% FCS group at 12 h and 24 h respectively. IL - 6 plus IL - Compared with 2% FCS group, the OD value and LI of PDGF group at 12 h and 24 h were both higher than that of 2% FCS group, but all experimental groups could not reach the cell proliferation level of 20% FCS group. The combination of two-factor stimulation 12 h and 24 h were no significant synergistic effect of promoting proliferation. Conclusion: Both IL - 6, IL - 1 and PDGF can stimulate the proliferation of rat glomerular Ms C to a certain degree. There is no obvious synergistic effect when combined with two - factor stimulation.
