本文报告5例4个月人胚(因妇产科指征小剖腹娩出)心肌细胞在培养前胰蛋白酶消化后培养1、2、3、4和5日后的透射电镜所见。胰蛋白酶消化对心肌细胞核、肌原纤维等都有明显影响,但培养24小时基本恢复正常。培养1、2和3日的人胚心肌细胞超微结构和培养前的心肌细胞基本相似。培养4和5日的部分心肌细胞出现 Z 线变形、肌节不完全等变性变化,部分心肌细胞仍具较正常结构。用培养的人胚心肌细胞制备病理模型,宜在培养后1、2和3日内进行。
This article reports 5 cases of 4-month human embryos (obstetrics and gynecology indications small cesarean delivery) of myocardial cells cultured before trypsinization 1, 2, 3, 4 and 5 days after transmission electron microscopy. Trypsin digestion on the myocardial cell nucleus, myofibril, etc. have a significant effect, but basically returned to normal after 24 hours of culture. The ultrastructures of human embryonic cardiomyocytes incubated for 1, 2, and 3 days were similar to pre-culture cardiomyocytes. Part of cultured cardiomyocytes cultured on day 4 and 5 showed Z-line deformation, incomplete sarcomere and other degeneration changes, some of the cardiomyocytes still had a more normal structure. The culture of human embryonic cardiomyocytes prepared pathological model should be carried out within 1, 2 and 3 days after culture.
