目的 观察肺纤维化大鼠支气管肺泡灌洗液(BALF)中干扰素γ(IFN γ)、白介素4(IL 4)的变化及卡介菌多糖核酸(BCG PSN)的调节作用。方法 采用成组设计,雄性SD大鼠随机分为正常对照组、博莱霉素(BLM)组和BCG PSN组,对苏木精伊红染色(HE)病理结果行半定量评价,分析BALF中细胞总数及分类;用酶联免疫吸附试验(ELISA)法检测BALF中IFN γ、IL 4 的含量。结果 ①和正常对照组(IFN γ:131.033±19.261,IL 4:7.155±1.001)比较,BLM组7天、14天BALF中IFN γ下降(58.167±9.227,86.022±10.997,P均<0.01),7天BALF中IL 4升高(11.15±1.415,P<0.01);②和BLM组比较,BCG PSN组IFN γ上升(BCG PSN7d:104.047±17.012,BCG PSN28d:171.667±29.930,P均<0.01),IL 4无下降(P>0.05);③BCG PSN组BALF中细胞总数及分类以及病理变化与BLM组比较,差异无显著性(P>0.05)。结论 ①在BLM致大鼠肺纤维化的BALF中,IFN γ含量减少,IL 4含量增加;②BCG PSN可促进IFN γ的分泌,部分纠正Th1/Th2细胞因子失衡。
Objective To observe the changes of interferon γ (IFN γ), interleukin 4 (IL 4) and the regulation of BCG PSN in bronchoalveolar lavage fluid (BALF) of rats with pulmonary fibrosis. Methods In a group design, male SD rats were randomly divided into normal control group, BLM group and BCG PSN group. The hematoxylin and eosin staining (HE) pathological results were semi-quantitatively evaluated. The total number of cells and their classification were determined. The levels of IFN-γ and IL-4 in BALF were detected by enzyme-linked immunosorbent assay (ELISA). Results ① In the BLM group, the IFNγ levels in BALF were decreased at 7 and 14 days (58.167 ± 9.227, 86.022 ± 10.997, P <0.01, respectively) compared with the control group (IFNγ: 131.033 ± 19.261 and IL 4: 7.155 ± 1.001) (P <0.01). (2) Compared with BLM group, IFNγ in BCG PSN group was increased (BCG PSN7d: 104.047 ± 17.012, BCG PSN28d: 171.667 ± 29.930, P <0.01) (P> 0.05). (3) There was no significant difference in the total number of cells and their pathological changes in BALF between BCG PSN group and BLM group (P> 0.05). Conclusions ① In BALF-induced pulmonary fibrosis in BALF, the content of IFN-γ and the content of IL-4 in BALF are increased; ② The secretion of IFN-γ is promoted by BCG PSN and the imbalance of Th1 / Th2 cytokines is partially corrected.
