目的利用PCR及测序方法分析青海地区回、汉族胎膜早破孕妇MMP-1、7、9基因多态性特点及是否具有民族差异性。方法提取胎膜早破孕妇和对照组组织DNA,利用PCR和琼脂糖凝胶电泳、测序法,检测基因多态性并行相关分析。结果 MMP-1 rs1799750 G/G、-/G、-/-基因型频率在汉族对照组和胎膜早破组中分别是44.25%、51.33%、4.42%和34.29%、47.14%、18.57%,两组基因型频率分布具有显著差异性(P=0.006);在回族对照组和胎膜早破组中基因型频率分别是45.98%、50.57%、3.45%和51.72%、34.48%、13.79%,两组基因型频率呈现显著差异性(P=0.005)。MMP-7 rs11568818三种基因型A/A、A/G、G/G在汉族对照组和胎膜早破组中基因型频率分别是60.18%、37.17%、2.65%和84.29%、8.57%、7.14%,两组相比,基因型频率分布具有显著差异性(P=0.000);回族对照组和胎膜早破基因型频率分布分别是9.77%、39.08%、1.15%和86.21%、6.90%、6.90%,两组基因型频率相比具有显著差异性(P=0.000)。MMP-9 rs3918242 SNP三种基因型C/C、C/T、T/T在汉族对照组和胎膜早破组中基因型频率分布分别是72.57%、26.55%、0.88%和70.00%、28.57%、1.43%,其基因型频率在两组中无显著性差异(P=0.895);在回族对照组和胎膜早破组中基因型频率分别是71.26%、8.74%、0.00%和70.69%、27.59%、1.72%,两组相比无显著差异性(P=0.468)。结论 MMP-1 rs1799750和MMP-7 rs11568818基因多态性可能与青海汉、回族胎膜早破相关;而MMP-9 rs3918242基因多态性与胎膜早破无关。同时,MMP-1rs1799750、MMP-7 rs11568818和MMP-9 rs3918242基因频率分布在青海汉、回族对照组和胎膜早破组中无特异性。
Objective To analyze the characteristics of MMP-1, 7 and 9 polymorphisms in pregnant women with preterm premature rupture of membranes and their ethnic differences in Qinghai area by PCR and sequencing. Methods The DNA of premature rupture of membranes in pregnant women and control group were extracted and analyzed by PCR and agarose gel electrophoresis and sequencing. Results The frequencies of genotypes of MMP-1 rs1799750 G / G, - / G, - / - were 44.25%, 51.33%, 4.42% and 34.29%, 47.14% and 18.57% respectively in the Han control group and premature rupture of membranes group, The frequencies of genotypes in the two groups were significantly different (P = 0.006). The frequencies of genotypes were 45.98%, 50.57%, 3.45% and 51.72%, 34.48% and 13.79% in the Hui control group and premature rupture of membranes group, The genotype frequency of the two groups showed significant difference (P = 0.005). The genotype frequencies of A / A, A / G and G / G of MMP-7 rs11568818 in Han control group and premature rupture of membranes group were 60.18%, 37.17%, 2.65% and 84.29%, 8.57% 7.14%, respectively. The frequency distribution of genotypes in Hui control group and premature rupture of membranes were 9.77%, 39.08%, 1.15% and 86.21%, 6.90% , 6.90%. There was significant difference between the two genotype frequencies (P = 0.000). The frequencies of genotypes C / C, C / T and T / T of the three genotypes of MMP-9 rs3918242 SNP were 72.57%, 26.55%, 0.88% and 70.00% in Han control group and premature rupture of membranes group respectively %, 1.43%, respectively. Conclusion The polymorphisms of MMP-1 rs1799750 and MMP-7 rs11568818 may be related to the premature rupture of membranes in Qinghai Han and Hui ethnicity. However, the polymorphism of MMP-9 rs3918242 is not associated with premature rupture of membranes. Meanwhile, the frequency distribution of MMP-1 rs1799750, MMP-7 rs11568818 and MMP-9 rs3918242 genes in Qinghai Han, Hui control group and premature rupture of membranes group was not specific.
