[目的]内生真菌代谢产物球毛壳菌素A具有优异、广谱的抑菌活性,具备良好的新型杀菌剂的开发前景。为了更好的分析发酵过程母液中球毛壳菌素A的含量,采用反相高效液相色谱法,建立了内生真菌代谢产物球毛壳菌素A定量分析方法。[结果]经梯度洗脱优化出的色谱条件:使用DuPont ZORBAX ODS C18色谱柱,以乙腈为流动相A、水为流动相B,梯度条件:(A)0~10 min,20%~25%;10~20 min,25%~40%;20~55 min,40%~55%;55~75 min,55%~75%;75~85 min,75%~90%。流速为1.0 mL/min,柱温:25℃,波长为254 nm。[结论]该方法精密度为1.49%,重现性为5.41%,可以作为球毛壳菌发酵优化过程中代谢产物球毛菌素A的检测技术。
[Objective] The product of endostar fungus GLA has broad spectrum antibacterial activity and possesses good development prospect of new bactericide. In order to better analyze the content of chaetocin A in the mother liquor of the fermentation process, a quantitative analysis method of the endochitinase metabolites, carbichostatin A, was established by reversed-phase high performance liquid chromatography. [Result] The chromatographic conditions optimized by gradient elution were as follows: using DuPont ZORBAX ODS C18 column with acetonitrile as mobile phase A and water as mobile phase B under gradient conditions: (A) 0 ~ 10 min, 20% ~ 25% ; 10-20 min, 25-40%; 20-55 min, 40-55%; 55-75 min, 55-75%; 75-85 min, 75-90%. The flow rate was 1.0 mL / min, the column temperature was 25 ℃ and the wavelength was 254 nm. [Conclusion] The precision of this method was 1.49% and the reproducibility was 5.41%, which could be used as the detection technology for the metabolites Gelactorin A in the fermentation process of Chaetomium globosum.
