目的探讨新疆地区维吾尔族、汉族健康人和脑梗死患者血浆纤维蛋白原Bβ(FgBβ)-455G/A位点基因多态性频率分布以及与血浆纤维蛋白原水平的关系。方法采用聚合酶链反应限制片段长度多态性(PCR-RFLP)方法对新疆地区156例缺血性卒中患者(维吾尔族63例,汉族93例)和143例的正常人(维吾尔族63例,汉族80例),进行FgBβ-455G/A位点单核苷酸多态性(SNP)分析,磁珠法测定血浆Fg水平。结果(1)新疆维吾尔族、汉族之间FgBβ-455G/A的基因型和等位基因频率分布差异无统计学意义(P>0.05)。(2)维吾尔族、汉族脑梗死组FgBβ-455G/A基因多态性和等位基因频率分布与对照组比较差异有统计学意义(P<0.05)。(3)脑梗死组血浆(fibrinogen,Fg)水平高于对照组,差异有统计学意义(P<0.05),维吾尔族、汉族之间Fg水平无统计学差异(P>0.05)。结论新疆地区维吾尔族和汉族脑梗死患者FgBβ-455G/A基因多态性存在差异,Fgβ启动区-455G/A多态性与脑梗死的发生有关联关系,-455G/A+AA是脑梗死的易感基因型,-455A是易感等位基因。
Objective To investigate the frequency distribution of plasma fibrinogen Bβ (FgBβ) -455G / A locus in Uighur and Han nationality healthy people and patients with cerebral infarction in Xinjiang and its relationship with plasma fibrinogen level. Methods A total of 156 ischemic stroke patients (63 Uygur and 93 Han nationality) and 143 normal controls (Xinjiang Uighur 63 cases, Han 80 cases), FgBβ-455G / A single nucleotide polymorphism (SNP) analysis, magnetic beads method for the determination of plasma Fg levels. Results (1) There was no significant difference in genotype and allele frequencies of FgBβ-455G / A between Xinjiang Uygur and Han (P> 0.05). (2) The FgBβ-455G / A gene polymorphism and allele frequency distribution in Uygur and Han nationality group were significantly different from those in control group (P <0.05). (3) The levels of fibrinogen (Fg) in cerebral infarction group were higher than those in control group (P <0.05). There was no significant difference in Fg level between Uygur and Han (P> 0.05). Conclusion The polymorphisms of FgBβ-455G / A in patients with Uygur and Han nationality cerebral infarction in Xinjiang are different. The -455G / A polymorphism of Fgβ promoter region is associated with the occurrence of cerebral infarction. Susceptibility genotype, -455A is a susceptible allele.
