目的优选东革阿里的提取纯化工艺。方法以宽缨酮Eurycomanone(EN)的转移率为考察指标,综合采用单因素实验和正交实验,通过对乙醇浓度、提取时间、溶剂用量、提取次数的考察,优选东革阿里合理的提取工艺;在静态吸附和解吸实验筛选树脂的基础上,结合单因素和正交实验优选东革阿里采用大孔树脂纯化的工艺。结果东革阿里的最佳提取工艺为:加20倍量水回流提取2次,第一次2h,第二次1h;最佳纯化工艺为:采用HPD100型大孔树脂,以70%饱吸附量上样,上样液质量浓度0.25 g生药·m L-1,上样流速3 BV·h-1,洗脱乙醇体积分数30%,洗脱流速3B V·h-1。结论优选出的工艺操作简单、稳定可行、重复性好,可推广于工业化生产应用。
Objective To optimize the extraction and purification process of Tongkat Ali. Methods Based on the transfer rate of Eurycomanone (EN), single factor experiments and orthogonal experiments were used to investigate the effects of ethanol concentration, extraction time, solvent dosage and the number of extraction. On the basis of static adsorption and desorption screening resin, combining with single factor and orthogonal experiment, Tongkat Ali was purified by macroporous resin. Results Tongkat Ali’s best extraction process: plus 20 times the amount of water reflux extraction 2 times, the first 2h, the second 1h; the best purification process: using HPD100 macroporous resin to 70% full capacity The samples were loaded with a mass concentration of 0.25 g crude drug · mL-1, a sample flow rate of 3 BV · h-1, an ethanol volume fraction of 30% and an elution flow rate of 3B V · h-1. Conclusion The optimized process is simple, stable and feasible with good repeatability and can be widely applied in industrial production.
