目的:探讨信号转导子及转录活化子3(STAT3)在食管鳞癌细胞系中的激活情况以及STAT3与其下游靶基因产物VEGF和Bcl-2的表达情况。方法:采用免疫细胞化学法和Westernblotting法检测两株食管鳞癌细胞系中STAT3及p-Stat3(STAT3蛋白的活化形式),VEGF和Bcl-2蛋白的表达情况,并采用凝胶电泳迁移率(elec-trophoreticmobilityshiftassay,EMSA)法检测两株食管鳞癌细胞核中STAT3与DNA的结合活性。结果:食管鳞癌细胞中存在激活的STAT3信号传导通路,通路蛋白STAT3、p-STAT3及其下游靶基因产物VEGF,Bcl-2在细胞中均有表达,蛋白在细胞核中具有较高的DNA结合活性。结论:两种食管鳞癌细胞系中均有组成性激活的STAT3信号通路,提示STAT3信号通路可能在食管鳞癌发生、发展中起重要作用。
AIM: To investigate the activation of signal transducer and activator of transcription 3 (STAT3) in esophageal squamous cell carcinoma and the expression of STAT3 and its downstream target genes VEGF and Bcl-2. Methods: The expressions of STAT3 and p-Stat3 (activated forms of STAT3), VEGF and Bcl-2 protein in esophageal squamous carcinoma cell lines were detected by immunocytochemistry and Western blotting. elec-trophoreticmobilityshiftassay, EMSA) was used to detect the binding activity of STAT3 to DNA in two esophageal squamous cell carcinoma nuclei. Results: STAT3 signal transduction pathway existed in esophageal squamous cell carcinoma cells, and the expression of pathway protein STAT3, p-STAT3 and its downstream target gene products VEGF and Bcl-2 were both expressed in the cells, and the protein had higher DNA binding in the nucleus active. CONCLUSION: STAT3 signaling pathway is constitutively activated in both esophageal squamous cell carcinoma cell lines, suggesting that STAT3 signaling pathway may play an important role in the development and progression of esophageal squamous cell carcinoma.
