目的:探讨ZEB2和E-cadherin mRNA和蛋白在鼻咽癌组织中的表达情况及其与鼻咽癌侵袭转移的关系。方法:应用荧光定量PCR和免疫组织化学方法检测ZEB2和E-cadherin在39例鼻咽癌和12例炎性鼻咽黏膜组织中的表达,分析ZEB2和E-cadherin表达与鼻咽癌患者临床病理参数之间的关系及两者的相关性。结果:ZEB2mRNA和蛋白在鼻咽癌中的表达水平均高于炎性鼻咽黏膜组织(P<0.05),ZEB2mRNA和蛋白的高表达均与肿瘤的N分级和临床分期有关(P<0.05),而与性别、年龄和T分级无相关性(P>0.05);E-cadherin mRNA和蛋白在鼻咽癌中的表达水平均低于炎性鼻咽黏膜组织(P<0.05),E-cadherin mRNA和蛋白的低表达均与肿瘤的N分级有关(P<0.05),而与性别、年龄、T分级和临床分期无相关性(P>0.05);ZEB2mRNA与Ecadherin mRNA呈负相关(r=-0.367,P<0.05),ZEB2蛋白与E-cadherin蛋白呈负相关(r=-0.322,P<0.05),均差异有统计学意义。结论:在鼻咽癌中ZEB2表达上调、E-cadherin表达下调,其表达呈明显负相关,并与鼻咽癌转移有关,可能机制与ZEB2下调E-cadherin的表达促进细胞EMT有关。
Objective: To investigate the expression of ZEB2 and E-cadherin mRNA and protein in nasopharyngeal carcinoma and its relationship with the invasion and metastasis of nasopharyngeal carcinoma. Methods: The expression of ZEB2 and E-cadherin in 39 cases of nasopharyngeal carcinoma and 12 cases of inflammatory nasopharyngeal mucosa were detected by real-time PCR and immunohistochemistry. The expressions of ZEB2 and E-cadherin in patients with nasopharyngeal carcinoma were analyzed. The relationship between the parameters and the correlation between the two. Results: The expression of ZEB2 mRNA and protein in nasopharyngeal carcinoma was higher than that in inflammatory nasopharyngeal mucosa (P <0.05). The high expression of ZEB2 mRNA and protein were correlated with the N stage and clinical stage (P <0.05) (P> 0.05). The expression of E-cadherin mRNA and protein in nasopharyngeal carcinoma was lower than that in inflammatory nasopharyngeal mucosa (P <0.05), and the expression of E-cadherin mRNA (P <0.05), but no correlation with gender, age, T grade and clinical stage (P> 0.05). ZEB2 mRNA was negatively correlated with Ecadherin mRNA (r = -0.367 , P <0.05). There was a negative correlation between ZEB2 protein and E-cadherin protein (r = -0.322, P <0.05). CONCLUSION: ZEB2 expression is up-regulated and E-cadherin expression is down-regulated in nasopharyngeal carcinoma. The expression of ZEB2 is significantly negatively correlated with the metastasis of NPC. The possible mechanism is that ZEB2 down-regulates the expression of E-cadherin and promotes EMT.
