目的研究Rho蛋白鸟苷酸交换因子11(ARHGEF11)基因R1467H多态性与多囊卵巢综合征(PCOS)及其胰岛素抵抗(IR)的关系。方法应用聚合酶链反应-限制性片段长度多态性技术对136例PCOS患者和117名健康对照者ARHGEF11基因R1467H位点进行基因分型;同时进行人体测量学和代谢指标的检测。结果在PCOS组和对照组中,ARHGEF11基因R1467H位点的基因型和等位基因频率分布差异无统计学意义(P>0.05);PCOS组GA/AA基因型的空腹胰岛素和HOMA2-IR水平明显高于GG基因型(P<0.05),对照组GA/AA基因型的空腹血糖显著高于GG基因型(P<0.05),这种统计学差异经年龄、体质量指数校正后依然存在(P<0.05)。结论 ARHGEF11基因R1467H多态性与PCOS的发病无关,但可能与PCOS患者IR抵抗有关。
Objective To investigate the relationship between the R1467H polymorphism of Rho protein guanylate 11 (ARHGEF11) gene and polycystic ovary syndrome (PCOS) and insulin resistance (IR). Methods The R1467H locus of ARHGEF11 gene in 136 patients with PCOS and 117 healthy controls was genotyped by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). The anthropometric and metabolic parameters were also tested. Results There was no significant difference in genotype and allele frequencies of R1467H in ARHGEF11 gene between PCOS group and control group (P> 0.05). Fasting insulin and HOMA2-IR levels of GA / AA genotype in PCOS group were significantly higher than those in PCOS group (P <0.05). The fasting blood glucose of GA / AA genotype in control group was significantly higher than that of GG genotype (P <0.05), and the statistical difference was still corrected after age and body mass index (P <0.05). Conclusion The R1467H polymorphism of ARHGEF11 gene is not related to the pathogenesis of PCOS, but may be related to IR resistance in PCOS patients.
