对4个玉米自交系(PH4CV、PH6WC、郑58和昌7-2)根系中的内生细菌进行分离和鉴定,为获得有益微生物资源奠定基础。通过形态学观察、生理生化特征鉴定及16S r DNA基因序列同源性分析和构建系统发育树等方法对分离菌株进行生物学鉴定,以确定其种属。共分离到23株内生细菌,鉴定为6个属,包括芽孢杆菌属(Bacillus sp.)、假单胞菌属(Pseudomonas sp.)、寡养单胞菌属(Stenotrophomonas sp.)、肠杆菌属(Enterobacter sp.)、贪噬菌属(Variovorax sp.)、鞘氨醇杆菌属(Sphingobacterium sp.),其中优势属为芽孢杆菌属。4个玉米自交系内生细菌的种类不同,细菌种类由多到少依次是PH6WC、郑58、PH4CV和昌7-2。通过对不同玉米自交系内生细菌的比较分析,发现内生细菌的种类和数量与玉米基因型相关。
The endophytic bacteria in the roots of four maize inbred lines (PH4CV, PH6WC, Zheng 58 and Chang 7-2) were isolated and identified, which laid the foundation for obtaining beneficial microbial resources. Through the morphological observation, the identification of physiological and biochemical characteristics and 16S rDNA sequence homology analysis and the establishment of phylogenetic tree and other methods of biological identification of isolates to determine their species. A total of 23 endophytic bacteria were isolated and identified as six genera, including Bacillus sp., Pseudomonas sp., Stenotrophomonas sp., Enterobacteriaceae, Enterobacter sp., Variovorax sp., Sphingobacterium sp., And the dominant genus is Bacillus. The endophytic bacteria of four maize inbred lines differed in their types, with bacterial species ranging from more to less followed by PH6WC, Zheng 58, PH4CV and Chang 7-2. Through comparative analysis of endophytic bacteria in different maize inbred lines, it was found that the type and quantity of endophytic bacteria are related to maize genotype.
