目的:比较避光与见光提取的枳术丸挥发油中主要成分含量的差异性。方法:采用水蒸气蒸馏法提取挥发油,挥发油提取器分别采用不包裹(见光)或者用黑布包裹(避光)提取相同时间。采用HPLC比较2种挥发油中苍术酮和白术内酯Ⅰ,Ⅱ,Ⅲ的含量差异,流动相水(A)-乙腈(B)梯度洗脱(0~22 min,55%~64%B;22~28 min,64%~64%B;28~48 min,64%~100%B),检测波长220,275 nm。结果:见光与避光提取的挥发油得率分别为1.03%和1.05%。见光提取的挥发油与避光提取的挥发油相比,苍术酮含量显著降低,分别为73.01,100.3 mg·g-1;白术内酯Ⅰ显著升高,分别为0.757 2,0.354 1mg·g-1;白术内酯Ⅱ差异不明显,分别为2.026,2.158 mg·g-1;白术内酯Ⅲ显著升高,分别为0.412 4,0.321 9 mg·g-1。结论:见光与避光提取的枳术丸挥发油总量无明显差异,但主要成分含量存在明显差异。提示在研究复方物质基础时,应注意实验过程中可能引起的内在成分变化。
OBJECTIVE: To compare the differences of the main components in the volatile oil of Zhizhu Pills extracted from light and light. Methods: Volatile oil was extracted by steam distillation. The volatile oil extractor used the same time without package (see light) or with black cloth (dark). HPLC method was used to compare the content of atractyloside and atractylenolide Ⅰ, Ⅱ and Ⅲ in two kinds of volatile oils. The mobile phase water (A) -acetonitrile gradient (0-22 min, 55% -64% B; 22 ~ 28 min, 64% -64% B; 28-48 min, 64% -100% B). The detection wavelength was 220,275 nm. Results: The yield of volatile oil extracted with light and light was 1.03% and 1.05% respectively. Compared with the volatile oil extracted by darkness, the content of atractylenol was significantly decreased by 73.01 and 100.3 mg · g-1, respectively, while that of Atractylodes macrocephala was significantly increased (0.757, 2.0354 and 1 mg · g -1, respectively) ; Atractyloside II was not significantly different, respectively 2.026, 2.158 mg · g-1; Atractylodes lactone Ⅲ increased significantly, respectively 0.412 4,0.321 9 mg · g-1. Conclusion: There is no significant difference in the total volatile oil of Zhizhu pill between light and darkness extraction, but there are significant differences in the main components. It is suggested that when studying the basis of compound materials, attention should be paid to the possible change of the intrinsic components in the experiment.
