目的初步探讨聚乙二醇(PEG)沉淀法对流感病毒气溶胶的浓缩效果。方法于2012年12月—2013年1月随机选取南京市5家商场超市和3家综合医院,采集室内可能含流感病毒的气溶胶样品,气溶胶经过不同的吸收液浓缩量、PEG-6000和NaCl加入量进行浓缩后提取RNA,以一步法逆转录巢式PCR检测甲型流感病毒。结果当PEG-6000和NaCl终浓度为7%和2.9%(m/V)浓缩10、20 ml吸收液以及PEG-6000和NaCl终浓度为14%和5.8%(m/V)浓缩10 ml吸收液时,甲型流感病毒的阳性检出率分别为17.5%,60%,30%,差异有统计学意义(P=0.008)。结论 PEG沉淀法可应用于流感病毒气溶胶的浓缩,并利于流感病毒的检出。
Objective To investigate the effect of polyethylene glycol (PEG) precipitation on the concentration of influenza virus aerosol. Methods From December 2012 to January 2013, five shopping malls and three general hospitals in Nanjing were randomly selected to collect aerosol samples that may contain influenza virus in the indoor. The aerosols were subjected to different concentrations of absorption liquid, PEG-6000 and RNA was concentrated to extract RNA, reverse transcription by one-step reverse transcription nested PCR detection of influenza A virus. Results Concentrations of 10 and 20 ml absorbent and PEG-6000 and NaCl concentration of 14% and 5.8% (m / V) concentrated 10 ml of PEG-6000 and NaCl at a final concentration of 7% and 2.9% (m / The positive detection rates of influenza A virus were 17.5%, 60% and 30% respectively, the difference was statistically significant (P = 0.008). Conclusion PEG precipitation method can be applied to the concentration of influenza virus aerosol, and conducive to the detection of influenza virus.
