目的研究D5S818、D8S1179、D7S820、CSF1PO、D2S1338、D3S1358、vWA、D21S11、D16S539、PentaE、TPOX、TH01、D19S433、D18S51、FGA、D6S1043、D13S317、D12S391等18个短串联重复序列(short tandem repeats,STR)基因座在甘肃地区东乡族人群的遗传多态性。方法采用PCR扩增及毛细管电泳技术对615名甘肃地区东乡族个体18个STR基因座进行分析。结果 615名甘肃地区东乡族个体在18个STR基因座上,共检出213种等位基因,850种基因型,其分布均符合Hardy-Weinberg平衡(P>0.05),18个基因座的杂合度(H)在0.633~0.928之间,匹配概率(Pm)在0.013~0.205之间,个体识别概率(DP)在0.795~0.987之间,多态信息含量(PIC)在0.560~0.920之间,非父排除概率(PE)值在0.332~0.854之间。结论实验研究了甘肃地区东乡族人群18个STR基因座的遗传多态性,为人类群体遗传学及法医学后续研究提供详实可靠的基础数据。
OBJECTIVE To study the effects of 18 short short tandem repeats (STRs) on D5S818, D8S1179, D7S820, CSF1PO, D2S1338, D3S1358, vWA, D21S11, D16S539, PentaE, TPOX, TH01, D19S433, D18S51, FGA, D6S1043, D13S317 and D12S391 ) Locus in Dongxiang, Gansu Province. Methods Twenty-eight STR loci of Dongxiang individuals in Gansu were analyzed by PCR and capillary electrophoresis. Results A total of 213 alleles and 850 genotypes were detected in 18 STR loci of 615 Dongxiang individuals in Gansu. The distributions of them were in accordance with Hardy-Weinberg equilibrium (P> 0.05), 18 loci (H) was between 0.633 and 0.928, the matching probability (Pm) was between 0.013 and 0.205, the individual identification probability (DP) was between 0.795 and 0.987, and the polymorphism information content (PIC) was between 0.560 and 0.920. The non-parent exclusion probability (PE) is between 0.332 and 0.854. Conclusions The genetic polymorphism of 18 STR loci in Dongxiang tribe in Gansu was studied experimentally, providing detailed and reliable basic data for the follow-up study of human population genetics and forensic science.
