目的:探讨补肾方剂对去卵巢大鼠骨丢失的疗效作用及其作用机理。方法:选用6月龄雌性SD大鼠,切除卵巢并诱发的骨质疏松模型,分别设为假手术组、模型组、雌二醇组和中药治疗组,治疗3个月后,观察大鼠骨密度、骨组织形态学及骨代谢相关生化指标。结果:补肾方剂组大鼠与模型组大鼠比较:补肾方剂可提高大鼠的平均骨密度,病理切片可见骨小梁明显致密,骨髓腔缩小。其生化指标显示雌二醇(E2)、黄体生成素(LH)、皮质醇(COR)、雌激素水平升高,而睾酮(T)水平明显降低,各指标之间统计分析均有统计学意义(P<0.05)。结论:补肾方剂有类似雌激素样作用,可有效防治卵巢切除大鼠的骨量丢失,可作为绝经后骨质疏松症的预防用药。
Objective: To investigate the effect and mechanism of Bushen Prescription on bone loss in ovariectomized rats. METHODS: Six-month-old female Sprague-Dawley rats were used to remove ovariectomized osteoporosis models. They were sham-operated group, model group, estradiol group, and traditional Chinese medicine treatment group. After 3 months of treatment, the rat bone was observed. Density, bone histomorphology, and biochemical markers related to bone metabolism. RESULTS: Compared with the model group rats, the formula of kidney-tonifying prescription can increase the average bone density of rats, and the pathological section shows that the trabecular bone is dense and the marrow cavity is reduced. The biochemical indicators showed that estradiol (E2), luteinizing hormone (LH), cortisol (COR), and estrogen levels were elevated, while testosterone (T) levels were significantly decreased. Statistical analysis was statistically significant among the indexes. (P<0.05). Conclusion: Kidney-tonifying formula has similar estrogen-like effects and can effectively prevent bone loss in ovariectomized rats. It can be used as a preventive drug for postmenopausal osteoporosis.
