水动力转染方法是指从小鼠尾静脉快速注射大体积含目的基因的生理盐水,从而实现外源基因在小鼠体内(尤其是肝脏内)的高效表达。它作为一种快速、简单、高效、安全的外源基因转染方法,已经广泛用于体内基因功能、基因调节、蛋白表达和基因治疗等方面的研究。由于该方法转染的目的基因主要在小鼠体内肝脏获得高水平表达,特别适用于肝炎病毒功能基因小鼠体内表达模型的建立。利用该技术建立的肝炎病毒实验动物模型有助于促进肝炎病毒致病机制的研究以及抗病毒药物的体内筛选与评价。
Hydrodynamic transfection refers to the rapid injection of a large volume of physiological saline containing the gene of interest from the tail vein of a mouse to achieve efficient expression of foreign genes in mice, especially in the liver. As a rapid, simple, efficient and safe method of exogenous gene transfection, it has been widely used in the study of gene function, gene regulation, protein expression and gene therapy in vivo. Since the target gene transfected by the method is mainly expressed in the liver of mice in vivo, it is particularly suitable for establishing the in vivo expression model of the hepatitis virus functional gene mouse. The experimental animal model of hepatitis virus established by the technology can help to promote the pathogenesis of hepatitis virus and in vivo screening and evaluation of antiviral drugs.
