目的探讨miR-126在人乳腺癌组织中的表达及其对人乳腺癌细胞(MCF-7和MDB-MA-231)增殖和侵袭的影响。方法收集2012年至2014年期间郑州大学附属肿瘤医院20例乳腺癌患者组织标本,realtime PCR检测乳腺癌组织和癌旁组织miR-126的表达。采用Lipofectamine TM2000将miR-126 mimics转染至MCF-7和MDA-MB-231细胞中,通过MTT实验检测MCF-7和MDA-MB-231细胞的增殖,Transwell法检测细胞的侵袭能力。结果乳腺癌标本中miR-126表达较癌旁正常组织中明显下调(P<0.05),miR-126的高表达明显的抑制了MCF-7和MDA-MB-231细胞株的侵袭性(P<0.05),但是在转染后经过48h的培养后,我们没有观察到细胞繁殖能力有明显改变。结论 miR-126在乳腺癌组织和细胞中低表达,miR-126抑制乳腺癌细胞株的侵袭性,但是对细胞繁殖能力没有明显影响。
Objective To investigate the expression of miR-126 in human breast cancer and its effect on the proliferation and invasion of human breast cancer cells (MCF-7 and MDB-MA-231). Methods Twenty cases of breast cancer tissues from 20 affiliated cancer hospitals in Zhengzhou University from 2012 to 2014 were collected. Real-time PCR was used to detect the expression of miR-126 in breast cancer tissues and adjacent tissues. The miR-126 mimics was transfected into MCF-7 and MDA-MB-231 cells with Lipofectamine TM2000. The proliferation of MCF-7 and MDA-MB-231 cells was detected by MTT assay. Results The expression of miR-126 in breast cancer specimens was significantly lower than that in normal tissues (P <0.05). The high expression of miR-126 significantly inhibited the invasiveness of MCF-7 and MDA-MB-231 cell lines (P < 0.05), but we did not observe significant changes in cell proliferation after 48 h of transfection. Conclusion miR-126 is low expressed in breast cancer tissues and cells, miR-126 inhibits the invasiveness of breast cancer cell lines, but has no significant effect on cell proliferation.
