目的 :检测MRP基因在人乳腺癌组织中的表达状况 ,初步分析其临床意义。方法 :37例待手术乳腺癌患者随机分为直接手术、术前化疗、术前化疗加三苯氧胺三组。采用RT -PCR法检测癌组织中MRP基因表达。结果 :术前化疗组MRP阳性率28.6 %(6/21) ;直接手术组MRP极弱或无表达(0/8) ;(P<0.01) ;加用三苯氧胺MRP表达减少为1/8。局部晚期病例MRP基因表达多呈阳性(5/9)。结论 :以上结果提示MRP是乳腺癌MDR产生的机制之一 ;MRP基因表达与乳腺癌的发展有关 ,可能是乳腺癌的重要预后指标
Objective : To detect the expression of MRP gene in human breast cancer and analyze its clinical significance. Methods: Thirty-seven breast cancer patients undergoing surgery were randomly divided into three groups: direct surgery, preoperative chemotherapy, and preoperative chemotherapy plus tamoxifen. RT-PCR was used to detect the expression of MRP gene in cancer tissue. Results: The positive rate of MRP in the preoperative chemotherapy group was 28.6% (6/21); the MRP in the direct surgery group was very weak or no expression (0/8); (P<0.01); and the expression of MRP was increased by 1/8 with tamoxifen. MRP gene expression was mostly positive in locally advanced cases (5/9). Conclusion: The above results suggest that MRP is one of the mechanisms of MDR in breast cancer; MRP gene expression is related to the development of breast cancer and may be an important prognostic indicator of breast cancer.
