低机械指数超声辐照SonoVue对脑缺血模型大鼠脑细胞凋亡诱导的影响

目的探讨低机械指数超声辐照SonoVue对脑缺血模型大鼠脑组织细胞凋亡诱导的影响。方法 SPF级成年Wistar大鼠56只,随机分为空白对照组10只,低剂量模型组12只、中剂量模型组12只、高剂量模型组12只和高剂量造影剂组10只,其中对照组和高剂量造影剂组脑供血正常,低、中、高剂量模型组结扎双侧颈动脉制作脑缺血模型。5组大鼠均行颅顶开窗术人造声窗,应用Logiq E9彩超仪进行超声辐照,探头ML6-15,频率15MHz,机械指数0.09,软组织热指数0.00,辐照时间20min。辐照开始时,低、中、高剂量模型组和高剂量造影剂组分别经尾静脉快速推注0.18、3.6、9.0、9.0mL/kg SonoVue溶液。5组大鼠辐照结束后行组织病理学检查,观察脑组织形态学变化,采用TUNEL法检测各组脑细胞凋亡情况。结果低、中、高剂量模型组大鼠端脑顶叶、侧叶和海马区可见不同程度缺血病灶,呈局灶性和/或弥漫性缺血改变;对照组和高剂量造影剂组未见明显形态学改变;低、中、高剂量模型组脑细胞凋亡指数(488.30±133.73、416.96±109.63、423.97±100.38)明显高于对照组(78.94±39.38)和高剂量造影剂组(126.97±64.91)(P<0.01),低、中、高剂量模型组脑细胞凋亡指数比较差异无统计学意义(P>0.05),对照组脑细胞凋亡指数与高剂量造影剂组比较差异无统计学意义(P>0.05)。结论低机械指数为0.09条件下SonoVue微泡在给药剂量9.0mL/kg范围内对脑组织细胞凋亡无明显促进作用。 Objective To investigate the effects of SonoVue irradiation induced by low mechanical index on cerebral cell apoptosis in rats with cerebral ischemia. Methods Fifty-six adult Wistar rats of SPF grade were randomly divided into blank control group (n = 10), low dose model group (n = 12), middle dose model group (n = 12), high dose model group (n = 12) and high dose contrast agent group Cerebral blood supply was normal in the group and high dose contrast media group. Cerebral ischemia models were made by ligating the bilateral carotid arteries in the low, medium and high dose model groups. All the rats in the 5 groups were given artificial cranial window. The Logiq E9 color doppler was used for ultrasound irradiation. The probe ML6-15, frequency 15MHz, mechanical index 0.09, soft tissue thermal index 0.00 and irradiation time 20min. At the beginning of irradiation, the low, medium and high dose model groups and the high dose contrast medium group were injected with 0.18, 3.6, 9.0 and 9.0 mL / kg SonoVue solution through tail vein respectively. The rats in the five groups were sacrificed and the histopathological examination was performed to observe the morphological changes of brain tissue. The apoptosis of brain cells in each group was detected by TUNEL method. Results Low, medium and high dose groups of rats showed ischemic lesions of varying degrees in the parietal lobe, lateral lobe and hippocampus, with focal and / or diffuse ischemic changes. The control group and high dose contrast agent group (488.30 ± 133.73, 416.96 ± 109.63, 423.97 ± 100.38) was significantly higher than that of the control group (78.94 ± 39.38) and the high dose contrast medium group (126.97 ± 64.91) (P <0.01). There was no significant difference in apoptotic index between low, medium and high dose groups (P> 0.05). There was no difference in apoptosis index between the control group and high dose contrast medium Statistical significance (P> 0.05). Conclusion SonoVue microbubbles had no significant effect on the apoptosis of brain cells at a dose of 9.0 mL / kg with a low mechanical index of 0.09.
