目的建立一种反胶束萃取L-辛弗林的方法,并验证利用P204/正辛醇萃取L-辛弗林的有效性。方法制备P204/正辛醇反胶束体系萃取枳实L-辛弗林,反萃取液经浓缩干燥后,获得纯化L-辛弗林。利用基于单因素实验进行的正交实验研究最佳分离工艺,通过紫外分析、原子力显微镜表征证实了P204/正辛醇反胶束体系萃取分离L-辛弗林的有效性。结果萃取最佳工艺条件为:水相pH值6.5、萃取时间15 min、P204浓度0.10mol/L、W_0=5,此条件下L-辛弗林的单次萃取率为68.02%,各影响因素影响顺序为:水相pH>P204浓度>W_0>萃取时间。纯化后的L-辛弗林纯度达到87.8%,比原料纯度提升了2.9倍。结论该方法为L-辛弗林的分离提取提供一种快速低成本方法,并对其萃取机制及主要变化规律方面提供了基础数据。
OBJECTIVE To establish a reverse micelle extraction method for L-synephrine and verify the effectiveness of L-synephrine extraction using P204 / n-octanol. Methods P204 / n-octanol reverse micellar system was used to extract the citrus aurantium L-synephrine, and the extract was concentrated and dried to obtain the purified L-synephrine. The optimal separation process was studied by orthogonal experiments based on single factor experiments. The UV spectroscopy and atomic force microscopy confirmed the effectiveness of the extraction and separation of L-synephrine by P204 / n-octanol reverse micelles system. Results The optimal extraction conditions were as follows: the pH value of aqueous phase was 6.5, the extraction time was 15 min, the concentration of P204 was 0.10 mol / L and W 0 was 5. Under this condition, the single extraction rate of L-synephrine was 68.02% The order of influence is: pH of aqueous phase> concentration of P204> W_0> extraction time. After purification, the purity of L-synephrine reached 87.8%, which was 2.9 times higher than the purity of raw materials. Conclusion This method provides a rapid and low cost method for the separation and extraction of L-synephrine, and provides the basic data for its extraction mechanism and main variation rules.
