目的观察FK506对丙烯酰胺(acrylamide,ACR)染毒导致的大鼠周围神经行为功能异常的影响。方法Wistar雄性大鼠60只,随机分为正常对照组、模型组、FK506低剂量组和高剂量组。模型组、FK506低和高剂量组分别给予ACR30mg/kg腹腔注射,正常对照组给予等体积生理盐水,1h后分别给予FK506皮下注射0.5和1mg/kg,5次/周,持续4周,每周步态评分并测量大鼠体重、甩尾指数、痛觉阈值、后肢撑力指数和转棒指数等指标。结果FK506低、高剂量组大鼠步态评分、后肢撑力指数明显低于模型组(第4周步态评分分别降低19.1%和43.4%,后肢撑力指数分别降低8.6%和31.3%),甩尾指数、痛觉阈值和转棒指数明显高于模型组(第4周甩尾指数分别升高20.4%和23.3%,痛觉阈值分别升高33.3%和38.5%),差异均有统计学意义(P<0.01),且FK506组间显示有剂量-效应依赖关系。结论FK506具有保护ACR导致的大鼠周围运动和感觉神经损伤的作用。
Objective To investigate the effect of FK506 on behavioral dysfunction of peripheral nerve in rats induced by acrylamide (ACR). Methods Sixty male Wistar rats were randomly divided into normal control group, model group, FK506 low dose group and high dose group. The model group, FK506 low and high dose groups were given ACR30mg / kg intraperitoneal injection, the normal control group given an equal volume of saline, 1h after FK506 subcutaneous injection of 0.5 and 1mg / kg, 5 times / week for 4 weeks, weekly Gait score and measurement of body weight, tail flick index, pain threshold, hindlimb bracing index and rod index and other indicators. Conclusion FK506 has the protective effect on ACR-induced peripheral motor and sensory nerve injury in rats.
