[目的]制备抗载脂蛋白apoAI鸡卵黄抗体(IgY),并研究其应答持久性、稳定性。[方法]用载脂蛋白apoAI免疫生蛋鸡,琼脂糖免疫扩散法测定IgY抗体效价变化。PEG法结合Sephadex-150凝胶过柱提取抗体;ELISA法测抗体对温度的稳定。[结果]免疫产蛋母鸡,末次免疫后其卵黄抗体最高滴度达到1︰128,Lowry法测IgY蛋白质浓度为8.11mg/L,70℃以下稳定性良好。[结论]用载脂蛋白apoAI免疫鸡制备高效价的特异抗体IgY,抗体应答时间持久、热稳定性好。
[Objective] To prepare anti-apolipoprotein apoAI chicken egg yolk antibody (IgY) and to study the persistence and stability of its response. [Method] Immunization of laying hens with apolipoprotein apoAI and the change of titer of IgY antibody by agarose immunodiffusion method. Antibodies were extracted by PEG method with Sephadex-150 gel and the temperature was stabilized by ELISA. [Result] The highest titer of yolk antibody reached 1: 128 after the last immunization and the IgY protein concentration was 8.11 mg / L by Lowry method. The stability under 70 ℃ was good. [Conclusion] Immunization of chickens with apolipoprotein apoAI to produce high titer of specific antibody IgY, antibody response lasting, thermal stability.
