目的 :探讨哮喘患者外周血单个核细胞(PBMC)IL 5、IL 3、GM CSFmRNA表达以及与慢性支气管炎患者的差异。方法 :选哮喘患者13例 (哮喘组 ) ,平均年龄(31.8±8.5)岁。慢性支气管炎患者9例 (慢支组 ) ,平均年龄(64.8±4.7)岁。健康献血员9例 (对照组 ) ,平均年龄(32.8±5.2)岁。抽取静脉血分离单个核细胞 ,用斑点印迹杂交法检测IL 5、GM CSF和IL 3mRNA的表达。结果 :哮喘组PBMCIL 5、GM CSF和IL 3mRNA表达的相对含量明显高于慢支组和对照组(P<0.01) ,慢支组与对照组相比无显著差异(P>0.05)。结论 :哮喘IL 5、GM CSF和IL 3mRNA表达明显增高 ,可能在哮喘发病中起重要作用。
Objective: To investigate the expression of IL-5, IL-3 and GM-CSF mRNA in peripheral blood mononuclear cells (PBMCs) of patients with asthma and their differences with chronic bronchitis patients. Methods: Thirteen asthmatic patients (asthma group) were selected, with an average age of (31.8 ± 8.5) years. Nine patients with chronic bronchitis (chronic bronchitis group), mean age (64.8 ± 4.7) years old. Nine healthy blood donors (control group), mean age (32.8 ± 5.2) years old. Venous blood was collected for the isolation of mononuclear cells and the expression of IL-5, GM-CSF and IL-3 mRNA was detected by dot blot hybridization. Results: The relative contents of PBMCIL 5, GM CSF and IL 3 mRNA in asthma group were significantly higher than those in chronic bronchitis group and control group (P <0.01). There was no significant difference between chronic bronchitis group and control group (P> 0.05). Conclusion: The expressions of IL-5, GM-CSF and IL-3 mRNA in asthma were significantly increased, which may play an important role in the pathogenesis of asthma.
