目的为了充分了解CO2安乐死装置的应用和进行动物安乐死的效果,比较动物种属和数量与死亡时间有无差异,为评价和使用CO2安乐死提供依据.方法用HOPE-MED8160型小动物善终处置设备对三种实验室常用的啮齿类小动物(大鼠、小鼠、豚鼠)进行CO2安乐死处理,向安乐死仓内缓慢充入CO2气体直到到达最高浓度99.9%,分别观察1只、5只、10只动物在进行CO2安乐死时达到死亡的CO2浓度,动物死亡所需要的时间,动物在死亡过程中的各种反应,包括神经行为改变、呼吸变化,无线遥测系统检测的心率、体温以及活动度的变化等.结果随着CO2浓度的增加,动物的呼吸先加快、加深后减慢,最终变得极其微弱直至死亡,心率逐渐减慢至0.大鼠和小鼠的死亡时间(分别是(4.80±0.64)min和(4.28±0.58)min)和死亡时CO2浓度(分别是(89.20±4.32)%和(87.33±2.52)%)之间无统计学差异(P>0.05),豚鼠死亡时间较长,(5.92±0.77)min,死亡时的CO2浓度达到99.9%.动物的心跳终止均是在呼吸停止后一小段时间(约0.5min).同时放置同种动物数量越多,死亡需要时间会有所延长.动物体温在整个安乐死过程中基本没变化,活动度在开始通CO2时有所增加,大约到30%~40%的CO2浓度时活动度变为0.结论 CO2小动物善终处置设备可以进行安乐死,动物在死亡过程中无明显的挣扎、抽搐现象.CO2处理是一种合适的动物安乐死方式.","Objective To observe the applications and the effects of euthanasia device with carbon dioxide(CO2) and compare the difference of death time on species and quantity,so as to provide basis for the evaluation and application of CO2 euthanasia.Method Three species of rodent experimental animals were caused death in euthanasia with CO2,including mice and guinea pigs,which were the most common animals used in laboratories.We used the HOPE-MED8160 small animal euthanasia equipment to process three rodent animals through filling with CO2 gas to the euthanasia chamber slowly until it reached the highest concentration of 99.9%.Observe one,five and ten animals respectively,including the CO2 concentration led to death,the death time,animals' reaction in the dying process,including changes of neurobehaviors and respirations.The changes of heart rates,temperature and activity will be detected and recorded by wireless telemetry system.Results With the increase of CO2 concentration,generally animals' breathing sped up and deepened at first,and then slowed down and eventually became extremely weak until death.Animals' heart rates slowed down to 0 gradually.The CO2 concentration was(89.20±4.32)% and(87.33±2.52)% on rats and mice respectively,causing the animals' death and the death time were(4.80±0.64)min and(4.28±0.58)min,which had no statistical significance(P<0.05).The death time(5.92±0.77)min of guinea pigs was longer,and the CO2 concentration to cause guinea pigs' death reached to 99.9%.Animals' heart rates always arrested a short time after the breathing stop(about 0.5min).In the meantime,the more the number of animals to place,the longer time for them to death.Animals' body temperature was constant during the euthanasia process.The activity of the animals at the start of filling CO2 increased and became 0 about 30%~40% of the CO2 concentration.Conclusions Small animal hospice disposal equipment with CO2 can be used for animal euthanasia.In the dying process,animals have no obvious struggle and twitch.Therefore CO2 treatment is a suitable way of animal euthanasia.
