目的:初步探讨海宁市结核分枝杆菌的数目可变串联重复序列(VNTR)的分型及分布特征,了解海宁市目前结核分枝杆菌的主要流行株,给结核病防治工作提供分子流行病学依据。方法:采用VNTR分型方法,提取DNA,采用PCR和琼脂糖凝胶电泳技术对结核分枝杆菌15个VNTR位点进行检测,并通过BioNumerics 5.0软件进行DNA指纹图谱多态性分析。结果:95株结核分枝杆菌的15个VNTR位点结果显示明显的多态性,经基因聚类分析,共分为10个基因群,共90个基因型。其中群占2.1%,群占1.1%,群占1.1%,群占5.3%,群占1.1%,群占1.1%,群占69.5%,群占1.1%,群占14.7%,群占3.2%。结论:初步证实海宁市2008年的95株结核分枝杆菌VNTRS存在明显的多态性,至少存在10个VNTR群,主要流行群为群。
OBJECTIVE: To investigate the classification and distribution of variable tandem repeat (VNTR) in Mycobacterium tuberculosis in Haining, and to find out the main epidemic strains of Mycobacterium tuberculosis in Haining and to provide a molecular epidemiological basis for the prevention and treatment of tuberculosis . Methods: The VNTR genotyping method was used to extract DNA. 15 VNTR sites of Mycobacterium tuberculosis were detected by PCR and agarose gel electrophoresis, and DNA fingerprinting polymorphism was analyzed by BioNumerics 5.0 software. Results: The 15 VNTR loci of 95 strains of Mycobacterium tuberculosis showed obvious polymorphism. According to the genetic clustering analysis, there were 10 gene groups with 90 genotypes. The group accounts for 2.1%, the group accounts for 1.1%, the group accounts for 1.1%, the group accounts for 5.3%, the group accounts for 1.1%, the group accounts for 1.1%, the group accounts for 69.5%, the group accounts for 1.1%, the group accounts for 14.7% and the group accounts for 3.2% . CONCLUSION: It is preliminarily confirmed that there are obvious polymorphisms in 95 strains of Mycobacterium tuberculosis VNTRS in Haining city in 2008, at least 10 VNTR groups exist, and the main epidemic groups are groups.
