小孢子胚转化成苗是油菜高效小孢子培养体系的重要环节。探索了基因型、激素、冷处理、琼脂糖浓度以及胚大小等因素对油菜小孢子胚成苗率的影响,结果表明:采用一定措施可明显提高胚成苗率,获得更多的小孢子苗。不同基因型材料的小孢子胚成苗率有明显不同,最高的胚成苗率可达42.9%;添加激素GA3和提高培养基的琼脂浓度至1.5%可提高部分基因型材料的成苗率;在4℃下处理4~7 d也可提高成苗率;同一基因型内长度>5 mm的小孢子胚成苗率较高。
Microspore embryo into seedlings is an important part of rape efficient microspore culture system. The effects of genotype, hormone, cold treatment, agarose concentration and embryo size on seedling rate of microspore embryo of rapeseed were explored. The results showed that embryo germination rate could be significantly increased and more microspore seedlings could be obtained. The microspore embryo germination rate of different genotypes was significantly different, the highest embryo germination rate was 42.9%. Adding hormone GA3 and increasing agar concentration to 1.5% could increase the seedling rate of some genotypes. At 4 ℃ for 4 ~ 7 d, the seedling rate could also be increased; the microspore embryo germination rate was longer in the same genotype> 5 mm.
