取豚鼠球囊、椭圆囊及壶腹嵴,置于含胶原酶的Hank's液中消化,再辅以机械分离方法获得单个离体Ⅰ型和Ⅱ型前庭毛细胞。Ⅰ型细胞为烧瓶状,有明显颈部、皮板及纤毛。Ⅱ型细胞较Ⅰ型细胞为短,无明显颈部,但有皮板及纤毛。Ⅰ型细胞与Ⅱ型细胞数量之比为12∶1。离体5小时后仍有35%的前庭毛细胞具有活性。观察结果表明,应用胶原酶能获得较多具有活性的单个离体前庭毛细胞,为前庭的生理、病理及药理实验研究提供了细胞模型。
Take guinea pig balloon, oval capsule and ampulla ridges, placed in Hank’s solution containing collagenase digestion, supplemented by mechanical separation of single isolated type Ⅰ and type Ⅱ vestibular hair cells. Type I cells are flask-shaped, with distinct neck, skin and cilia. Type II cells than Type I cells are short, no obvious neck, but there are skin and cilia. The ratio of type I cells to type II cells is 12: 1. 35% of the vestibular hair cells remain active after 5 hours in vitro. The results show that the application of collagenase to obtain more active single vestibular hair cells in vitro, for vestibular physiological, pathological and pharmacological experimental study provided a cell model.
