目的:探讨薏仁对糖尿病肾病大鼠肾组织中β1整合素及黏着斑激酶(FA K )表达的影响。方法50只大鼠随机均分为正常对照组(A组)、糖尿病肾病模型组(B组)、薏仁治疗组(C组)、厄贝沙坦治疗组(D组)和薏仁+厄贝沙坦联合治疗组(E组),C、D、E组分别以薏仁400 mg?kg‐1?d‐1、厄贝沙坦20 mg?kg‐1?d‐1、薏仁400 mg?kg‐1?d‐1+厄贝沙坦20 mg?kg‐1?d‐1水剂灌胃治疗至8周末。采用HE染色观察大鼠肾组织病理变化,qRT‐PCR和Western blot分别检测β1整合素、FAK的mRNA及其蛋白表达。结果 B 组β1整合素及 FAK 的 mRNA 和蛋白表达均较 A 组增加(P<0.05);与B组相比,C、D、E组β1整合素及FAK的mRNA和蛋白表达降低(P<0.05);与C组相比,E组β1整合素及FAK的mRNA和蛋白表达略降低(P>0.05)。结论薏仁通过下调肾组织中β1整合素及FA K的表达,从而达到减轻肾小球纤维化、延缓糖尿病肾病进展的作用。“,”Objective To investigate the effect of coix seed on the expressions of β1‐integrin and focal adhesion kinase(FAK) in the renal tissues of rats with diabetic nephropathy (DN) .Methods Fifty rats were equally randomized into five groups of A (normal controls) ,B(DN model) ,C(treated with coix seed 400 mg?kg‐1 ?d‐1 ) ,D(treated with irbesartan 20 mg?kg‐1 ?d‐1 ) and E(treated with coix seed 400 mg ? kg‐1 ? d‐1 plus irbesartan 20 mg ? kg‐1 ? d‐1 ) .The pathological changes of renal tissues were observed by HE staining .The mRNA and protein expressions of β1‐integrin and FAK were detected by qRT‐PCR and Western blot ,respectively .Results The mRNA and protein expressions of β1‐integrin and FAK in group B were higher than those in group A ( P<0 .05 ) . Compared with group B ,the mRNA and protein expressions of β1‐integrin and FAK were decreased in groups of C ,D and E ( P0 .05) .Conclusion Coix seed can protect renal function and delay DN progress probably via downregulating the expressions ofβ1‐integrin and FAK in the renal tissues .
