目的:评价复方中药“天芪航力片”对大鼠正加速度(+Gz)致心肾损伤的防护效果,为减轻高G损伤提出新的防护措施。方法:将36只雄性SD大鼠随机分为3组(每组12只),即对照组(A组)、+Gz组(B组)和实验药物组(C组)。C组连续给予复方中药“天芪航力片”提取物14 d,每天1次,给药量为20 ml/kg。A组和B组每天给予等量的蒸馏水。14 d后,B组和C组进行+10 Gz持续5 min,增长率为1 G/s的高G暴露。各组均于+Gz作用后1 h,采集血样分离血清,送第四军医大学西京医院临床免疫科测定大鼠血清心脏损伤标志物肌酸激酶(CK)和肌酸激酶同工酶(CK-MB),以及肾脏损伤标志物血清尿素氮(BUN)和肌酐(Cr)的含量。对肾脏ATP酶的表达进行组织化学染色法检测。取肾脏组织超薄切片进行电镜观察。结果:B组血清CK和CK-MB以及C组的CK-MB含量较A组均显著升高(P<0.01),而C组CK的含量较A组显著升高(P<0.05)。B组大鼠血清BUN的含量较A组显著降低(P<0.05),血清Cr的含量显著升高(P<0.05)。C组血清BUN及Cr的含量与A组比较无显著变化,但与B组相比BUN含量有显著性差异(P<0.05)。B组肾脏ATP酶阳性细胞数较A组显著降低,而C组ATP酶阳性细胞数则无明显变化。B组肾脏超微结构出现明显损伤性改变而C组则无明显变化。结论:复方中药“天芪航力片”提取物对+Gz引起的心肾损伤具有一定的防护作用。
Objective: To evaluate the protective effect of Compound Chinese Medicine Tianqi Hanglidian on heart and kidney injury induced by positive acceleration (+ Gz) in rats, and to provide new protective measures to reduce high G damage. Methods: Thirty-six male Sprague-Dawley rats were randomly divided into 3 groups (12 in each group): control group (group A), + Gz group (group B) and experimental drug group (group C). Group C was continuously given compound Chinese medicine “Tianqi Air Force tablets” extract 14 d, day 1, the dose was 20 ml / kg. Groups A and B were given equal amounts of distilled water daily. After 14 days, groups B and C performed a high G exposure of +10 Gz for 5 min with an increase of 1 G / s. Blood samples were collected at 1 h after + Gz treatment, and serums were collected and serum creatine kinase (CK) and creatine kinase (CK-MB) were measured in clinical immunology department of Xijing Hospital, Fourth Military Medical University. MB), and the levels of serum urea nitrogen (BUN) and creatinine (Cr) as markers of kidney injury. The renal ATPase expression was detected by histochemical staining. Take ultra-thin sections of kidney tissue under electron microscope. Results: The contents of CK-MB in group B and CK-MB in group C were significantly higher than those in group A (P <0.01), while those in group C were significantly higher than those in group A (P <0.05). The level of serum BUN in group B was significantly lower than that in group A (P <0.05), and the content of serum Cr was significantly increased (P <0.05). The levels of serum BUN and Cr in group C had no significant changes compared with those in group A, but the levels of BUN in group C were significantly different (P <0.05). The number of ATPase positive cells in group B was significantly lower than that in group A, while the number of ATPase positive cells in group C did not change significantly. The ultrastructural changes of renal ultrastructure in group B were obviously changed, but in group C there was no obvious change. Conclusion: The compound traditional Chinese medicine “Tianqi Air Force Tablets” extract has a protective effect on + Gz induced heart and kidney injury.
