目的:研究金属硫蛋白(MT)对缺氧和复氧豚鼠乳头状肌动作电位的影响.方法:利用标准的玻璃微电极技术,将标本暴露于缺氧液(低氧,高钾,无糖,pH68)和MT.结果:MT(002mmol·L-1)对正常豚鼠乳头状肌的动作电位无影响;单纯将标本暴露于缺氧液,明显使APD20,APD50和APD90缩短,增高RP水平,降低APA及Vmax;缺氧液加入MT(002mmol·L-1)后,则使缺氧期间的RP,APA和Vmax的改变恢复正常,但APD进一步缩短;同时发现MT减少复氧期间导致的自律性的发生率.结论:MT具有钙调节剂的特征.
Objective: To investigate the effect of metallothionein (MT) on the action potentials of papillary muscles in hypoxia and reoxygenation guinea pigs. METHODS: Specimens were exposed to hypoxic solutions (hypoxia, high potassium, sugarless, pH 6.8) and MT using standard glass microelectrode techniques. Results: MT (002 mmol·L-1) had no effect on the action potential of papillary muscles in normal guinea pigs. Exposure of the specimens to hypoxic solution alone significantly shortened APD20, APD50 and APD90, increased RP levels and decreased APA and Vmax (P <0.05). After adding hypoxic solution (0.02 mmol·L-1), the changes of RP, APA and Vmax in hypoxia returned to normal, but the APD was further shortened. At the same time, MT was found to reduce the autonomic Occurrence rate. Conclusion: MT has the characteristics of calcium regulators.
