目的调查广西凭祥地区莱姆病感染情况和基因型别。方法 2011年7月从广西凭祥地区采集蜱、啮齿动物和野鸟标本,分别采取煮沸法和Qiagen试剂盒提取蜱、啮齿动物脾脏以及鸟脾脏中莱姆病螺旋体基因组DNA;采用巢式PCR扩增莱姆病螺旋体5S~23SrRNA基因间隔区;对PCR扩增产物进行测序,并将序列结果与GenBank中莱姆病螺旋体5S~23SrRNA基因间隔区序列比对分析。结果从3份啮齿动物标本中检测到莱姆病螺旋体5S~23SrRNA基因间隔区片段,啮齿动物的感染率为5.66%(3/53)。其中一个序列与GenBank中Borrelia valaisiana基因型莱姆病螺旋体(序列号:HM100125.1,AB091455.1,AB091454.1,AB091453.1)同源性为100%;蜱和鸟标本中未检测到莱姆病螺旋体。结论广西凭祥地区啮齿动物中存在莱姆病螺旋体B.valaisiana基因型感染。
Objective To investigate the Lyme disease and genotype in Pingxiang, Guangxi. Methods Ticks of ticks, rodents and wild birds were collected from Pingxiang, Guangxi province in July 2011. The genomic DNAs of ticks, rodent spleens and spleen of Borrelia burgdorferi were extracted by boiling method and Qiagen kit respectively. PCR amplification products were sequenced, and the sequence results were compared with the 5S ~ 23SrRNA spacer sequences of Lyme disease spirochetes in GenBank. Results The 5S ~ 23S rRNA gene spacer region of Borrelia burgdorferi was detected from 3 rodent specimens. The rodent infection rate was 5.66% (3/53). One of these sequences shared 100% identity with Borrelia valaisiana genus Borrelia burgdorferi (sequence numbers HM100125.1, AB091455.1, AB091454.1, AB091453.1) in GenBank. Spirochete Conclusion The B. valaisiana genotype is present in rodents in Pingxiang, Guangxi.
