目的分析载脂蛋白E(ApoE)基因多态性及其对慢性乙型肝炎病毒(HBV)感染性免疫的影响。方法白细胞介素(IL)-2检测采用放射免疫法,聚合酶链反应(PCR)法扩增ApoE基因片段,用荧光偏振的方法检测ApoE基因型。结果轻度组ApoEε4高于正常值范围,并与中度、重度组相比差异有统计学意义(P均<0.05),与ApoEε3、ApoEε2组相比ApoEε4组有较高的IL-2和总胆固醇水平。结论高水平的ApoEε4基因可通过调节IL-2和血脂水平减轻HBV感染后引起的肝细胞炎症反应及肝损伤。
Objective To analyze the polymorphism of apolipoprotein E (ApoE) gene and its effect on the infectious immunity of chronic hepatitis B virus (HBV). Methods Detection of interleukin - 2 (IL - 2) ApoE gene fragment was amplified by radioimmunoassay and polymerase chain reaction (PCR), and ApoE genotypes were detected by fluorescence polarization assay. Results ApoEε4 in mild group was higher than normal, and there was significant difference compared with moderate and severe group (all P <0.05). Compared with ApoEε3 and ApoEε2 group, ApoEε4 group had higher IL-2 and total Cholesterol level. Conclusions ApoEε4 gene of high level can alleviate the hepatocellular inflammatory response and liver injury caused by HBV infection by regulating IL-2 and blood lipid levels.
